Adenovirus-mediated gene expression in the septal cells of cirrhotic rat livers

Abstract

We previously reported that in rat liver fibrosis induced by carbon tetrachloride (CCl4), the adenovirus-mediated gene transfer to hepatocytes was markedly reduced compared with normal rat liver. Moreover, in cirrhosis there was even a stronger reduction of the number of transduced hepatocytes; the recombinant adenovirus vectors would transfer a foreign gene to the septal cells of cirrhotic rat livers. These results raised a question regarding the infectivity and localization of adenovirus vector in the cirrhotic rat. In this study, therefore the infectivity of the vectors in vitro was examined using slice cultures from the cirrhotic rat livers. Furthermore, the localization of adenovirus were estimatied in the kidney which is the second target organ of adenovirus. As a result, in vitro infection to the slice culture demonstrated that the cirrhotic hepatocytes still maintained the infectivity of adenovirus. In addition, in the kidney the infectivity was observed no differences between normal and cirrhotic rats. In conclusion, the septal cells in the cirrhotic liver should be targeted by the in vivo gene transfer with the adenovirus vector.
