Invited Symposium: Molecular Mechanisms of Ageing

TIG-3, a normal human diploid fibroblast line established in our institute, shows the finite life span around 70-80 population doubling levels (PDL). The cell line was used for prepairing a 2-D gel protein database directed to researches on cellular aging mechanisms. Cellular proteins were resolved by two-dimensional gel electrophoresis in combination of immobilized pH-gradient isoelectric focusing and vertical gel slab SDS-PAGE. The clickable 2-D gel protein map was prepared by silver staining, since most of the post-translationally modified proteins which might play an important role in cellular aging process are eliminated from the map if that is prepared by radioactive methionine labeling.
