Ischemia-Reperfusion induced Changes in Levels of Ion Transport Proteins in the Gerbil Brain

Abstract

Derangement of ion homeostasis is proposed as critical step responsible for delayed neuronal death of hippocampal neurons after ischemia-reprfusion injury (IRI). A quantitative Western blot was used to asses the levels of ion transport translation products in control and in animals after IRI. The plasma membrane Ca-pump(PMCA) is is a Ca high affinity ubiquitously expressed protein used along with of organellar Ca pump(SERCA) as the main determinats for lowering cytosolic Ca in hippocampal neurons.The IP3 receptor type I is the most abundant in the cerebellum and hippocampus.On the response of external signal it releases Ca from internal stores. The Na pump is the enzyme responsible for the major portion of brain energy expenditute and the Na gradients. Antibodies against PMCA isoform were from dr. Guerini, Zurich, against alpha 3 Na pump from dr.Presley, Dallas and against IP3 receptor from dr.Parys, KUL.Immediatelly after isothermic ischemic insult in halothane anesthesia and or after 10 days after the operation, the male gerbils were decapitated.The gene products of PMCA showed distinct distribution pattern.In hippocampus are the most abundant PMCA1 and in a lesser amount PMCA3.The IRI selectively down-regulates the levels of PMCA1 protein and IP3 receptor type I. No statistically significant changes have been detected in the levels of SERCA 2b and reticular calreticulin as well as alpha 3 Na pump.We suppose that alteration of number of ion transport proteins can contribute to changes which participate and/or follow the delayed death of neurons in hippocampus.
