Characterization of Quinpirole-Stimulated [35S] GTPgammaS Binding at D2-like Receptors in Rat Striatum.

Materials and Methods

Quinpirole-stimulated [35S]GTPgammaS binding was assessed in striatal membranes prepared from adult male Sprague Dawley rats. Each assay tube contained agonist and/or test drug(s), 300 microM GDP, 0.1 nM [35S]GTPgammaS and membrane homogenate (40 - 60 micrograms protein), in a final buffer concentration of 20mM Hepes, 100mM NaCl, and 3mM MgCl2, pH 7.4. Reaction tubes were incubated at 37o C for 20 minutes, then terminated by rapid vacuum filtration. [35S]GTPgammaS binding was quantified by scintillation spectroscopy. Data are presented as percent stimulation over basal. The EC50, IC50, and Emax values were calculated by nonlinear regression analysis using a four parameter model (Sigma Plot, SPSS, Chicago, IL).
