Aging and Resistance to Oxidative Damage in Caenorhabditis elegans

Abstract

Much attention has focused on the role that oxidative damage plays in cellular and organismal aging. The C. elegans gene mev-1 confers resistance to oxidative stress and modulates longevity. We have cloned mev-1 by transformation rescue and found that it encodes cyt-1 encoding the large subunit of succinate dehydrogenase cytochrome b. A missense mutation abolishes complex II activity in the mitochondrial membrane. These data indicate that CYT-1 participates to mediate electron transfer. Moreover, mutational inactivation of this process renders animals susceptible to oxidative stress and as a result, leads to precocious aging.
