To Bernd Nilius: Your results with expression of Trp3 in CPAE cells are most exciting in view of the recent findings of Kiselyov et al. (Nature 396:478) demonstrating that hTrp3 provides a structure that is able to sense the activated IP3 receptor. Thus, one interpretation of your results would be that expression of the Trp3 isoform enables functional coupling of Trp channels to activated IP3 receptors in CPAE. Which other Trp isoforms are expressed in CPAE?  What are the requirements regarding intracellular free Ca in terms of a co-factor for IP3-induced activation of NSC in Trp3 transfected cells? Is this Cai dependence similar to that in EA cells?
