Multiple negative regulatory elements in 5'-flanking region of the rat catalase gene.

Abstract

Catalase activity is markedly decreased in malignant hepatoma cell lines, whereas enriched in normal liver cells. Previously, we found negative regulatory element located in -3269 to -1701 bp upstream of ATG and other multiple sites of positive- and negative regulatory elements in this region. In particular, a negative regulatory element at -2799 to -2652 bp was associated with some protein complexes with chracteristic configuration in hepatoma cells. This element was demonstrated to have strong silencer activity by CAT assay. The core sequence of this element was found in multiple sites widely within -3269 to -1701. These elements and the binding proteins suggested involving in strong repression of the catalase gene in hepatoma cells.
